So I'm about to have a ton of TDPs to spend (via circling) and I'm looking for the stat layout that will provide me with the most benefit for trading on routes and commodities. I currently have 33 Discipline and 49 Charisma. I can either do 33 Disc 75 Charisma or 50 Disc 65ish Charisma...What is optimal?

I try to keep my trader balanced for combat as well as trading- but if I were you, I would use the latter set-up, especially if you should get attacked on the trade routes.

My combats are so pathetic that if something more vicious than a Goblin attacks me on the trade routes I will be running for the hills, sorry Caravan Driver but you're on your own. But then if I'd wanted to fight things I'd be a Barbarian or something!

For just purely gaining money from routes, is Charisma the only contributing stat?

If anything, train some stamina so you can hold all those shiny coins without falling over. I fancy myself a swordsman, but it does help with the gruelling day-to-day caravan runs.

Pretty sure it is strength that helps you carry your masses of wealth (in bronze so that Thieves will be disappointed), and I already have 30 of it, which was pretty spendy for an Elothean!

It's a combination of Strength and Stamina.

In fact, it is both (Strength and Stamina) and are both weighted equally.

Carrying capacity in stones:

10 * [ 2*STRENGTH + 2*STAMINA + Floor([STRENGTH+STAMINA+4]/5) + Floor([STRENGTH+STAMINA+2]/5) ]

That formula, assuming it is correct, is much simplified thus:

10 * floor(2.4*STR + 2.4*STA + 0.8)

I should get around to including it on elanthipedia as a note under the original or something...

To address the original question, I'd put it all in charisma. Although as an Elothean, I'd be training mentals up high as well.
